Toremifene: Dosage, Mechanism Onset of Action, Half-Life
Ten patients on anticonvulsants (phenobarbital, clonazepam, phenytoin, and carbamazepine) showed a twofold increase in clearance and a decrease in the elimination half-life of toremifene. In a study of 20 healthy subjects, 2 mg midazolam once daily (days 6 and 18) coadministered with toremifene as a 480 mg loading dose followed by 80 mg once daily for 16 days. Following coadministration on days 6 and 18 relevant increases in midazolam and α-hydroxymidazolam Cmax and AUC were not observed.
- In multicentre comparative trials, objective responses (complete and partial) occurred in 20 to 29% of patients treated with toremifene (60 to 240 mg/day) and in 19 to 37.5% of tamoxifen (20 or 40 mg/day) recipients.
- The cause of the serum enzyme elevations during toremifene therapy is unknown, but they may represent fatty liver disease induced by the estrogen receptor modulation.
- This page contains brief information about toremifene and a collection of links to more information about the use of this drug, research results, and ongoing clinical trials.
- Unlike tamoxifen, toremifene has not been approved as a means of prevention of de novo breast cancer in high risk patients or for prevention of recurrence of breast cancer.

Toremifene is used in postmenopausal women to treat breast cancer. It is usually used to treat cancer that needs estrogen, a hormone, in order to grow (estrogen-receptor positive). Toremifene is a nonsteroidal antiestrogen that blocks the effects of estrogen in the breast tissue, thereby slowing or stopping the growth of cancer. Toremifene is used to treat metastatic breast cancer (cancer that has spread) that is hormone-receptor positive in women who have already stopped menstruating (postmenopausal). Sometimes it is not safe to use certain medications at the same time.
